This stack will enhance all important facets of acute cognitive enhancement

- Memory/Learning (AChE, AMPA)
- Motivation/Focus (Dopamine + NE)
- Energy/Wakefulness (Adenosine Inhib.)

Let's dive right into it
4
1) Citicoline (CDP Choline or Cognizin)
Dose: 500-1000mg daily

Citicoline is my go-to acetylcholine (ACh) precursor for daily use

Citicoline breaks into choline & uridine, which both boost ACh levels

And in case you're new, ACh is involved in learning, memory, & attention

5
Besides boosting ACh levels, Citicoline also enhances:

- Dopamine & NE levels (mild)
- Cell membrane formation & repair
- Brain blood flow

The result?

It reliably enhances various aspects of memory, reaction time, focus, attention, & more in human studies

2) Alpha GPC
Dose: 300-600mg (as needed)

Many nootropic enthusiasts believe Alpha GPC is the best ACh precursor

And they're not necessarily wrong

But Alpha GPC goes one step further
Media image
7
It's not only a precursor, but also seems to powerfully release acetylcholine stores

This is partially why it seems to yield such a potent acute effect after ingestion

Due to its potency & TMAO risks, I use this only as needed and instead take Citicoline daily
8
Nearly all Alpha GPC products are only 50% Alpha GPC, which was surprising to learn

I've had the best results with Toniiq's Alpha GPC, which is one of the few 99% Alpha GPC products on the market

3) Huperzine A
Dose: 100-200mcg (as needed)

Huperzine prevents the breakdown of acetylcholine by inhibitng acetylcholinesterase (AChE)

Unlike many other AChE inhibitors, Huperzine is selective for the G4 isoform

This is crucial as it reduces counterproductive side effects
Media image
10
AChE inhibitors are so cognitive-enhancing that they're used to treat Alzheimer's & dementia (e.g. Donepezil)

Donepezil is superior, but comes with additional risks

Huperzine is sufficient for most

Start with 100mcg until you know how you react

4) Nicotine or Tropisetron
Dose: 3mg (zyn) or 5-10mg (Trop)

Whether you like it or not, nicotine is cognitive-enhancing at 2-4mg

This is primarily due to its activation of the α7 nicotinic cholinergic receptor

But it also binds to the α4β2 receptor, which is addictive

12
Enter Tropisetron

"Nicotine without addiction"

It's an α7 nicotinic receptor partial agonist being investigated for the treatment of Alzheimer's

Early studies suggest Tropisetron is a potent cognitive-enhancer without addiction

13
Tropisetron also blocks the 5-HT3 receptor which further enhances cognition via releasing ACh

This action also reduces nausea and Tropisetron has long been used for this purpose

If sticking to nicotine, I would use it very sparingly as it's very easy to get addicted

5) Piracetam
Dose: 1600mg (up to 3x)

Piracetam is famous for being the first "nootropic"

It's a positive allosteric modulator of AMPA receptors

This increases the likelihood that information processing neurons fire electrical signals

The result is improved learning & memory
Media image
15
Besides its direct effect on AMPA receptors, it also upregulates:

- ACh receptors
- Brain energy metabolism
- Membrane fluidity

Its effect is mild, but well-documented & synergistic when used in this stack

If you're looking for Piracetam on steroids, consider TAK-653

16
TAK-653 is a very potent AMPA potentiator with minimal side effects due to its selecitivity

In initial human studies, it improved executive function and exerted antidepressant effects in a rat study

It's very much a research chemical, so use at your own risk

Dose = 2mg

6) Notable Focus
Dose: 2-4 capsules (5 days/wk)

I formulated this one because existing supplemental dopamine enhancers did NOTHING for me

Notable Focus is different...

It contains a powerful flavonoid called Oroxylin A, one of the only natural dopamine reuptake inhibitors
Media image
18
Aside from Oroxylin A, the other ingredients work to:

- Enhance dopamine synthesis
- Prevent dopamine breakdown
- Enhance DA sensitivity via neurogenesis

7) Adenosine Blockers
Dose: Varies

Caffeine has been shown time and time again to enhance cognition

It works primarily by blocking adenosine receptors

Much of the cognitve effects are specifically due to blocking A1 & A2A
Media image
Media image
20
Blocking A1 indirectly stimulates the release of neurotransmitters such as norepinephrine, acetylcholine, & others

Blocking A2A increases stimulation of dopamine D2 receptors due to the heteromeric complex formed by the two

To summarize, here's the recommended stack:

- Citicoline = 2x 500mg
- Alpha GPC = 300mg (as needed)
- Huperzine A = 100-200mcg (as needed)
- Nicotine = 3mg (sparingly as needed)
- Piracetam = 1600mg (up to 3x/day)
- Notable Focus = 2-4 caps
- Caffeine (or similar)

There are ton of others that could be included here, which I'll be discussing in future threads
