It examined giving:

◇ Glycine: an amino acid in foods like collagen

&

◇ N-Acetylcysteine: a modified form of the dietary amino acid cysteine

To older adults.

Both at a dose of 100 mg / kg bodyweight, daily, pretty big doses of both.

Older adults were randomly put in either the glycine + NAC (GlyNAC), or a placebo (alanine).

The study was 12 weeks.

They also had younger adults enroll to see if the supplementation could really make them biologically resemble youth.

Supplementing glycine + NAC restored every measure back to youthful levels.

Older adults saw:

◇ ↑ Muscle-glutathione - the body's master antioxidant
◇ ↑ Red blood cell (RBC)-total glutathione (tGSH)
◇ ↑ RBC-reduced glutathione (rGSH)
◇ ↑ RBC-GSH/GSSG ratio - the amount of "usable" vs "used" glutathione
◇ ↓ Plasma TBARS - a marker of oxidative damage
◇ ↓ Plasma F2-isoprostanes - a marker of lipid peroxidation (also oxidative damage)
◇ ↓ Muscle respiratory quotient (RQ) - an indicator of increased fat burning
◇ ↑ Muscle mitochondrial fatty-acid oxidation
◇ ↓ Muscle mitochondrial glucose oxidation
◇ ↑ Rate of palmitate (a common fat) oxidation

ALL of these markers improved to the point of being statistically indistinguishable from the young adults.

Wow.

Increases in:

◇ GCLC - helps form glutathione
◇ GSS - also helps form glutathione
◇ PGC1α - master mitochondrial biogenesis protein
◇ Sirt3 - activates mitochondrial energy production
◇ CPT1b - helps break down fat for energy
◇ PPARα - master fat burning controller
◇ HADHA - an enzyme in fat burning
◇ Complex I
◇ Complex II

◇ Complex V - all crucial proteins involved in mitochondrial energy production
◇ PINK1 - stimulates mitochondrial autophagy
◇ GLUT4 - helps bring glucose into cells for energy

NAC + glycine improves various markers of inflammation + insulin resistance, too.

Old adults completely reversed age related changes in:

➜ IL-6 – Inflammatory cytokine
➜ CRP – Systemic inflammation marker
➜ IL-10 – Anti-inflammatory cytokine
➜ 8-OHdG – Marker of DNA damage
➜ sICAM-1 – Endothelial activation marker
➜ sVCAM-1 – Vascular endothelial inflammation
➜ HOMA-IR – Insulin resistance index
➜ Insulin

While TNFα, another inflammatory cytokine, also declined, just not back to youthful levels.

Blood sugar and glucose tolerance were the only measures that did not improve.

Taken together, this has to be one of the most successful studies I have EVER seen in terms of improving multiple key markers.

You'd be hard pressed to find examples of anything showing all of these improvements, and to this degree.

But HOW is this combination so effective?

Both of these nutrients synergize to help form GLUTATHIONE - the body's master antioxidant.

Glutathione serves a central role in the antioxidant system:

➜ Recycles vitamins E and C
➜ Helps detoxify hydrogen peroxide - a key reactive oxygen species (ROS) molecule

Our mitochondria generate lots of ROS as a part of generating energy,

and their accumulation:

➜ Signals cells to stop taking in / burning energy
➜ Damages every component of the cell
➜ Triggers the inflammatory response
➜ Alters gene expression

This all results in impaired mitochondrial function, protein expression characteristic of aging, impaired insulin sensitivity, and even worsened body composition.

Essentially, this accumulation of ROS (oxidative stress or oxidative damage) is perhaps THE fundamental driver of the aging process.

NAC and glycine have both been studied independently to have benefits for related conditions,

but adding them together might even be greater than the sum of their parts.
